MS Medium: Applications in Plant Tissue Culture

Sep 16,2026

Applications

MS medium (Murashige and Skoog medium) is one of the most widely used basal media in plant tissue culture. It is commonly used for plant micropropagation, callus culture, organ culture, embryo culture, and cell suspension culture.

Depending on the plant species and culture objective, the standard MS formulation can be modified by adjusting nutrient concentrations or adding plant growth regulators such as 6-BA, NAA, and IBA. Both solid and liquid forms are used, with the choice depending on the type of culture being carried out.

1. Plant Micropropagation

MS medium provides the mineral nutrients required for the in vitro growth of many plant explants, including shoot tips, embryos, stem segments, and anthers. With an appropriate combination of plant growth regulators, it can support shoot multiplication, rooting, and other stages of plant regeneration.

It is therefore widely used in the propagation of vegetables, fruit trees, ornamental plants, and medicinal plants, particularly where uniform plant material is required.

2. Plant Breeding and Genetic Research

MS medium is also commonly used as a basal medium in plant breeding, genetic transformation, and in vitro selection. It provides a controlled culture environment for the regeneration and growth of plant tissues following different experimental treatments.

The nutrient composition and plant growth regulator combination can be adjusted according to the plant species and the specific regeneration system.

3. Germplasm Conservation

In vitro culture systems based on MS medium can be used in the maintenance and conservation of plant germplasm. This approach is useful for certain rare, specialty, or vegetatively propagated plants where conventional field or greenhouse maintenance may be difficult.

For long-term or slow-growth conservation, however, the culture medium and environmental conditions need to be specifically adjusted according to the plant material and conservation method.

4. Modified MS Media for Different Culture Requirements

The standard MS formulation is not suitable for every plant or every stage of tissue culture without modification. Reduced-strength formulations, such as 1/2 MS medium, are commonly used for certain culture stages or plant species.

The concentrations of macronutrients, micronutrients, vitamins, and plant growth regulators can be adjusted to meet specific requirements. This flexibility makes MS medium useful across a wide range of tissue culture applications, including micropropagation, plant regeneration, and the production of virus-free planting material.
